Assessment of Leukocytes

Elevated level of WBC in the semen is indicative of genital tract infections.
Presence of WBC in semen is also associated with the production of ROS (Residual Oxidative Stress), affecting sperm parameters like motility, morphology & DNA integrity.

Granulocytes WBC contain the peroxidase enzyme and detection of peroxidase positive WBCs is an important indicator of acute infection & inflammation. The detection of peroxidase enzyme on wet mount semen smear is simple, quick & reliable.

Note :

  • Differentiation of immature sperms & WBCs in the wet mount semen smear under phase contrast microscope is difficult.  Just by counting ‘Round Cells’, one cannot ascertain the presence of the WBCs.
  • Assessment of WBCs concentration by peroxidase – positive counts is well comparable with the WBC counts evaluated by more sophisticated technologies like Immunohistology and flow cytometery.
